It is highly unlikely that this is a new registered account.This project aims to understand the mechanisms by which aging affects biological function in the human central nervous system (CNS) by examining the morphology, neurochemistry, physiology and pharmacology of CNS cells from brains of aged subjects. Since degenerative changes in the human brain begin to appear very early in life, we are also studying these phenomena in primates to determine if they occur at the same time and same place. Since many of the more commonly used laboratory animal species are not relevant to human pathophysiology, we are also studying non-human primate models of human brain aging. Our recent studies suggest that the dendritic arbor of cerebellar Purkinje cells decreases markedly with age, but that the number of nerve cells per unit volume of tissue is unchanged. This finding suggests that the possible pathophysiological sequelae of this neuronal loss may be increased susceptibility to epileptic seizures, which develop more frequently in the elderly. To address this question, we have begun studies of the mechanisms responsible for the aging-related decrease in cell number, the arrangement of the nerve cells in the cerebellum, and the role of the glial cells. The increased susceptibility of the elderly to epilepsy appears to be related to the increased permeability of the blood brain barrier (BBB).
